This study tested the validity of the AmpFlSTR Yfiler PCR amplification kit in amplifying male DNA, with attention to the amplification of male DNA in the presence of a high background of female DNA, which is often a circumstance in sexual assault cases.
Abstract
Using FBI/National Standards and the guidelines recommended by the Scientific Working Group on DNA Analysis Methods (SWGDAM), the kit succeeded in verifying the best reaction conditions and reagent concentrations for the amplification of male DNA at low levels (below 125 pg) and the specific amplification of male DNA in the presence of a high background of female DNA (1:1,000). Under optimal conditions, no detectable cross-reactive products were obtained on human female DNA, bacteria, and commonly encountered animal species. The performance criteria included overall peak heights, intracolor peak balance, and lack of cross-reactive peaks in the presence of female DNA. The kit simultaneously amplifies 17 Y-STR loci, including the loci in the "European minimal haplotype" (DYS19, DYS3854a/b, DYS389I, DYS389II, DYS390, DYS391, DYS392, and DYS393); the SWGDAM recommended Y-STR loci (DYS438 and DYS439); and the highly polymorphic loci DYS437, DYS448, DYS456m DYS458, Y GATA H4, and DYS635. 5 tables, 8 figures, and 42 references
